RESUMO

To investigate the relationship between serum uric acid level and glomerular ischemic lesions (GIL) in patients with primary membranous nephropathy (PMN) and identify relevant risk factors. A total of 201 patients with PMN but normal renal function confirmed by renal biopsy executed in the Liaocheng People's Hospital, China, during January 2020-January 2023 were analyzed retrospectively. The enrolled patients were divided into a hyperuricemia group and a normal serum uric acid group (control group) according to their serum uric acid levels. Then, the participants were further divided into a non-GIL group or a GIL group based on the patient's renal biopsy results. The two groups' clinical and pathological data and meaningful indicators for differences were analyzed by binary logistic regression analysis. Additionally, the serum uric acid level prediction value on GIL was investigated using receiver operating characteristic (ROC) curves. Compared with the control group, the hyperuricemia group exhibited high serum uric acid, the prevalence of GIL, serum albumin, the prevalence of hypertension, and low-density lipoprotein cholesterol (LDL) levels (P < 0.05). Compared with the non-GIL group, the GIL group exhibited were older, had enhanced serum uric acid, serum albumin, and an increased prevalence of tubular atrophy/interstitial fibrosis (TA/IF), arteriolosclerosis, and low eGFR levels (P < 0.05). The binary logistic regression analysis revealed that the serum uric acid and the TA/IF are independent risk factors of GIL (P < 0.05). The AUC of ROC of GIL of PMN patients, predicted based on the serum uric acid concentration, was 0.736 (P < 0.05), wherein the threshold = 426.5 µmol/L and the Youden's index = 0.41. Serum uric acid concentration and the TA/IF are independent risk factors of GIL in patients with PMN, and the former exhibits prediction value on GIL in patients with PMN.

Contact resistance has become one of the main bottlenecks that hinder further improvement of mobility and integration density of organic field-effect transistors (OFETs). Much progress has been made in reducing contact resistance by modifying the electrode/semiconductor interface and decreasing the crystal thickness, however, the development of new organic semiconductor materials with low contact resistance still faces many challenges. Here, 2,6-bis-phenylethynyl-anthracene (BPEA) is found, which is a material that combines high mobility with low contact resistance. Single-crystal BEPA OFETs with a thickness of ≈20 nm demonstrated high mobility of 4.52 cm2  V-1  s-1 , contact resistance as low as 335 Ω cm, and band-like charge transport behavior. The calculated compatibility of the EHOMO of BPEA with the work function of the Au electrode, and the decreased |EHOMO -ΦAu | with the increase of external electric field intensity from source to gate both contributed to the efficient charge injection and small contact resistance. More intriguingly, p-type BPEA as a buffer layer can effectively reduce the contact resistance, improve the mobility, and meanwhile inhibit the double-slope electrical behavior of p-channel 2,6-diphenyl anthracene (DPA) single-crystal OFETs.

The retronasal aroma of Baijiu is closely related to its quality and consumer preference. Retronasal detection thresholds (RDTs) of 44 aroma compounds were determined in 46% v/v ethanol using a three-alternative forced-choice procedure, which varied widely and ranged from less than 0.02 to over 1,000,000 µg/L. Nineteen aroma compounds, including ß-phenylethanol, 2,3,5,6-tetramethylpyrazine, dimethyl trisulfide, and 2-methyl-3-(methyldisulfanyl)furan, had RDTs lower than their orthonasal detection thresholds. Power function curves were used to establish correlations between RDTs and partition coefficients for five esters and four pyrazines (R2  = 0.9575, 0.9969, respectively). Twenty-nine aroma compounds had retronasal odor activity values >1 in a soy sauce aroma type Baijiu. Additionally, time-intensity (TI) results suggested that hexanoic acid, ethyl hexanoate, isoamyl acetate, 3-methyl-butanal, and nonanal contributed to a "burst-aroma" of Baijiu. Conversely, TI results attributed the "after-odor" of Baijiu to dimethyl trisulfide, methional, 2,3,5,6-tetramethylpyrazine, ß-phenylethanol, and other compounds. PRACTICAL APPLICATION: This manuscript provides comprehensive information on the retronasal sensory characteristics of aroma compounds in Baijiu. The results obtained may help understand the contribution of aroma compounds to retronasal aroma perception during Baijiu tasting and give helpful information to the Baijiu industry regarding quality control.

This study aims to present retronasal aroma perception and oral aroma release of Baijiu by progressive profiling and intra-oral solid-phase microextraction (SPME) combined with comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (GC × GC-TOFMS). Fruity, alcohol, grain, and cellar were the active retronasal aroma attributes of the strong aroma type Baijiu S, and soy sauce, burnt, and roasted were attributes of the soy sauce aroma type Baijiu SS. The dynamic release behaviors of 91 oral aroma compounds of two Baijiu samples were characterized and classified into four or five groups of different oral persistence. Principal component analysis showed that vapor pressure and Henry constant were negatively correlated with the oral persistence of aroma compounds, of which the cumulative interpretation rate was 71.67%. Partial least squares regression analysis showed that aroma compounds, such as ethyl benzeneacetate, ß-phenylethanol, and tetramethylpyrazine, were related to the long-persistence retronasal aroma of Baijiu (Q2 = 0.806).

Background: For acquired immunodeficiency syndrome (AIDS) patients with suspected opportunistic infections, the rapid and accurate identification of pathogens remains a challenge. Metagenomic next-generation sequencing (mNGS) has emerged as a pan-pathogen assay for infectious diseases diagnosis, but its guiding significance for diagnosis and antimicrobials treatment in AIDS patients with suspected opportunistic infections is still not well established. In this study, we compared the microbiological diagnostic value of mNGS with that of conventional microbiological tests (CMTs) in AIDS patients with suspected opportunistic infections. Methods: From January 2018 to February 2021, a retrospective study was performed at four tertiary teaching hospitals in China and data of 86 AIDS patients with suspected opportunistic infections were collected. The pathogen detection performance of mNGS and CMTs were compared. Results: Positive agreement between mNGS and clinical diagnosis was significantly higher than that of CMTs (65/86 (75.6%) vs 37/86 (43.0%)). In addition, mNGS identified more bacterial (25 vs 2), fungal (5 vs 3), viral (9 vs 2) organisms compared with CMTs. Mixed infection were detected in 34 patients by mNGS combined with CMTs. Viruses (94.1%, 32/34) and fungi (94.1%, 32/34) were commonly seen in the mixed infection cases. mNGS helped identify the pathogen or guide appropriate treatment in 49/86 (57%) patients. Meanwhile, CMTs also contributed in the decision of appropriate treatment in 28 patients. The successful de-escalation or discontinuation of treatment was supported in 37 patients with the help of mNGS. We observed a significant reduction in the number of patients being prescribed foscarnet (52.3% vs 23.26%, p < 0.001), moxifloxacin (34.9% vs 10.5%, p = 0.005), and levofloxacin (32.6% vs 14%, p = 0.001) before and after mNGS. Conclusion: For AIDS patients with suspected opportunistic infections, mNGS can provide early, noninvasive, and rapid microbiological diagnosis. mNGS may lead to a more precise antimicrobial treatment and reduced the unreasonable use of antimicrobials.

OBJECTIVE: We retrospectively analyzed risk factors on in-hospital mortality in CRRT-therapy patients with open cardiac surgery (CS)-induced acute kidney injury (AKI), to provide the clinical basis for predicting and lowering the in-hospital mortality after CS. METHODS: 84 CS-AKI patients with CRRT were divided into survival and death groups according to discharge status, and the perioperative data were analyzed with R version 4.0.2. RESULTS: There were significant differences between the two groups, including: urea nitrogen, Sequential Organ Failure Assessment (SOFA) score and vasoactive-inotropic score (VIS) on the first day after operation; VIS just before CRRT; SOFA score and negative balance of blood volume 24 h after CRRT; the incidence rate of bleeding, severe infection and MODS after operation; and the interval between AKI and CRRT. Univariate logistic regression analysis showed that SOFA score and VIS on the first day after operation; VIS just before CRRT; VIS and negative balance of blood volume 24 h after CRRT; the incidence rate of bleeding, infection and multiple organ dysfunction syndrome (MODS) after operation; bootstrap resampling analysis showed that SOFA score and VIS 24 h after CRRT, as well as the incidence of bleeding after operation were the independent risk factors. CONCLUSION: Maintaining stable hemodynamics and active prevention of bleeding are expected to decrease the in-hospital mortality.

BACKGROUND: To investigate the relationship between serum uric acid levels and glomerular ischemic lesions in patients with immunoglobulin A nephropathy (IgAN) and the relevant risk factors. METHODS: A total of 86 patients with IgAN and normal renal functions were divided into a hyperuricemia group and a normal serum uric acid group (control group). These patients were further divided into a glomerular ischemic lesions group and a non-glomerular ischemic lesions group (control group) based on the renal biopsy results. The relationship between serum uric acid levels and glomerular ischemic lesions was analysed. RESULTS: In patients with IgAN, the prevalence or occurrence of glomerular ischemic lesions was significantly higher in the hyperuricemia group compared with the normal serum uric acid group. Elevated serum uric acid levels are independently associated with glomerular ischemic disease. CONCLUSION: Hyperuricemia in patients with IgAN may lead to glomerular ischemic lesions, and lowering serum uric acid levels may delay the progression of IgAN.

The ''after-odor" is essential to the quality and consumer preference of Baijiu. An intra-oral solid-phase microextraction (SPME) combined with comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (GC × GC-TOFMS) was developed for oral aroma compounds monitoring of Baijiu. The extraction time, sip volume and rinse time were 120 s, 5 mL, and 10 s, respectively. The procedure showed good performances at concentrations of 1.56 µg/L-1500 mg/L for different aroma compounds of Baijiu (most R2 > 0.9). Furthermore, the optimized procedure detected 85 aroma compounds with different chemical structures and provided good representations of the light, strong, soy sauce and mix aroma type Baijiu's oral aroma profiles. Principal component analysis's (PCA) cumulative interpretation rate was 72.42%. The acceptable performance of this procedure can be combined with dynamic sensory evaluation to improve the quality of the Baijiu aroma.

Organic single-crystalline semiconductors are highly desired for the fabrication of integrated electronic circuits, yet their uniform growth and efficient patterning is a huge challenge. Here, a general solution procedure named the "soft-template-assisted-assembly method" is developed to prepare centimeter-scale monolayer molecular crystal (MMC) arrays with precise regulation over their size and location via a capillary-confinement crystallization process. It is remarkable that the field-effect mobility of the array is highly uniform, with variation less than 4.4%, which demonstrates the most uniform organic single-crystal arrays ever reported so far. Simulations based on fluid dynamics are carried out to understand the function mechanism of this method. Thanks to the ultrasmooth crystalline orientation surface of MMCs, high-quality p-n heterojunction arrays can be prepared by weak epitaxy growth of n-type material atop the MMC. The p-n heterojunction field-effect transistors show ambipolar characteristics and the corresponding inverters constructed by these heterojunctions exhibit a competitive gain of 155. This work provides a general strategy to realize the preparation and application of logic complementary circuits based on patterned organic single crystals.

Hydrogen sulfide (H2S), a recognized environmental pollutant, comes from a wide range of sources. For example, H2S will be produced in the process of plant protein corruption, the decomposition of domestic sewage and garbage, food processing (wine brewing), etc. and once the concentration is too high, it will cause significant damage of environment and human body. Besides H2S is an important gas signal molecule in vivo, which can be transferred through lipid membrane. Its existence level is closely related to many diseases. If we can "visually" trace the transmembrane transmission of hydrogen sulfide, it will be very helpful for the study of oxidative stress processes, cell protection, signal transduction and related diseases closely related to H2S. Although some probes can detect H2S in environment, cytoplasm and organelles, there are few reports on the release and internalization of H2S. In this work, we report a H2S fluorescence probe that can retain on the cell membrane, named PCM. The probe PCM can not only detect endogenous and exogenous H2S, but also distinguish them, this provides a general strategy for the construction of probes to detect other biomarkers. In addition, PCM has been successfully applied to the detection of endogenous and exogenous H2S in zebrafish, which has the potential to become a new chemical tool and provide help for the research of H2S-related diseases.

Mercury is a highly toxic metal element, and the accumulation of mercury in the human body can cause great harm, including but not limited to brain damage, kidney damage and behavioral disorders. Therefore, an effective way to detect mercury ions in the environment is urgently needed. In this study, a novel fluorescent probe (CP-Hg) was synthesized with coumarin as the fluorophore and propanethiol as the recognition receptor. The probe was characterized with high sensitivity (detection limit is approximately 0.5 nM) and selectivity. Note that the probe can react with mercury ions with a distinct color change. In addition, it has been proved to have low toxicity and successfully applied to detect mercury in water samples, macrophages and zebrafish model.

Mercury, as a highly toxic heavy metal, can cause very serious harm to human health and even death in severe cases. Therefore, we synthesized a novel ratiometric fluorescent probe for detecting mercury ions, with mercaptoethanol as the recognition receptor. Probe CMER could determine mercury ions in 0-1.6 µM and the detection limit is 7.6 nM. Moreover, CMER manifested a fast response for Hg2+ (within 5 s) and simultaneously observed that the color changed from light yellow to orange by naked eye. In addition to these preeminent spectral properties, the probe also had satisfactory bioimaging results in RAW 264.7 macrophage cells and zebrafish.

Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+ ) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+ . After addition of Hg2+ , a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 µM and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

We have synthesized a simple Golgi-targeting H2S fluorescent probe which can detect endogenous and exogenous H2S in cells and zebrafish. In addition, this probe provides a new chemical tool for the detailed study of generation pathways of H2S under Golgi stress response.

Small molecular biothiols, such as cysteine (Cys), homocysteine (Hcy), reduced glutathione (GSH), and hydrogen sulfide (H2S), play crucial parts in regulating the redox balance of life activities, regulating normal physiological activities and preventing various diseases. Quantitative analysis of these important small molecular substances is very important for revealing their diverse physiological and pathological effects. Although many fluorescent probes have been reported to detect biothiols in cells, it is still not sufficiently advanced to detect biothiols with separated fluorescence emission peak by same wavelength excitation. In our work, we designed a simple conjugate of Nile red and NBD (7-nitro-1,2,3-benzoxadiazole) as long-wavelength fluorescent probe NR-NBD for the simultaneous discrimination of these biothiols at single wavelength excitation. Probe NR-NBD could efficiently discriminate Cys/Hcy, GSH and H2S by two separated fluorescence emission channels and absorption spectra. Importantly, probe NR-NBD has excellent specificity and sensitivity towards the monitoring of endogenous/exogenous Cys/Hcy and GSH/H2S in living cells and zebrafish.

As a eukaryotic organelle, the Golgi apparatus plays an essential role in various physiological activities such as stress response. The Golgi stress response is an important physiological process of conferring cytoprotection by regulating the synthesis and metabolism of bioactive molecules. Therefore, the development of new suitable in situ analytical techniques for monitoring related small molecular substances in the stress reaction of the Golgi apparatus is very helpful for further study of the regulatory mechanism of the Golgi apparatus. Recent studies have shown that endogenous hydrogen sulfide (H2S) also possesses crucial bioregulatory and protective performances in the stress response. Therefore, the high-fidelity in situ mapping of H2S production under the Golgi stress response plays an important role not only in revealing cytoprotection functions of H2S in the stress response but also in further understanding the regulatory mechanism of the Golgi stress response. In this work, we designed a simple Golgi-targetable H2S fluorescent probe (Gol-H2S) that responds accurately and sensitively to H2S in the Golgi apparatus of living cells and zebrafish. On the basis of its superior bioimaging performances, probe Gol-H2S was successfully applied to the in situ visualization of H2S production under the Golgi stress response elicited by monensin, a specific-Golgi stressor. The related process of the Golgi stress response was validated by stimulation and inhibition experiments. These findings fully demonstrate that H2S is an alternative biomarker of the Golgi stress response. Moreover, probe Gol-H2S can also be used as a potential tool for disclosing the detailed H2S-cytoprotection mechanisms under the regulation of the Golgi stress response in related diseases.

In recent years, targeting drugs made by physical loading or chemical bonding of drugs on small molecular carriers have shown a very wide application prospect in the field of tumor and cancer treatment. How to achieve the release of drugs in cancer cells has become the core of this research. One of the most important bases for drug localization is to use the difference of small molecular biothiol concentration between cancer cells and normal cells. Details of the changes of biothiol levels in the growth and reproduction of cancer cells are still poorly understood, and the main reason is the lack of sensitive real-time imaging tools for biothiols in cancer cells. In this work, we reasonably designed and synthesized the combination of 4-hydroxy-1,8-naphthalimide and NBD-Cl as a concise fluorescent probe HN-NBD for imaging biothiols in live cells and zebrafish. In addition, due to the advantages of HN-NBD design, it is sufficiently sensitive to biothiols, and further imaging can distinguish cancer cells from normal cells. Probe HN-NBD would be of great significance to biomedical researchers for the study of biothiol-related diseases, the screening of new anticancer drugs, and the early diagnosis and treatment of cancers.

Hypochlorous acid (HOCl) has received special attention by virtue of its pivotal antimicrobial nature, and the appropriate amount of HOCl is beneficial to innate immunity of host to cope with microbial invasion. However, the uncontrollable accumulation of HOCl is implicated in many human diseases and even cancers. Thus, to determine its deeper biological functions, it is significantly important to specifically monitor intracellular HOCl in biosystems. Herein, we rationally designed a simple fluorescent probe FH-HA on the basis of the formylhydrazine recognition receptor and rhodamine B fluorophore. It is worth noting that the formylhydrazine moiety for the first time is adopted as the recognition receptor for specifically recognizing HOCl. Additionally, probe FH-HA also exhibited excellent performance in many areas including satisfactory water-solubility, high specificity, and excellent sensitivity. Notably, probe FH-HA could quickly respond to HOCl (within 3 s), which facilitates the tracing of transient HOCl. More importantly, probe FH-HA was capable of specifically tracing the fluctuations of endogenous HOCl in living cells and zebrafish, and it could monitor basal HOCl in cancer cells to distinguish cancer cells from normal ones.

Mercury ions as high toxic pollutants have received wide-spread attention because of their poisonousness, persistence and enrichment. To better understand the distribution of mercury species and supplement more detailed toxicological research, it is necessary to develop some methods for monitoring mercury ions with high sensitivity and selectivity. Therefore, a simple rhodol-based highly selective fluorescent probe, RH-Hg, has been developed for monitoring Hg2+ with thiocarbamate as the recognition receptor. The probe RH-Hg can quantificationally detect mercury ions in aqueous solution assisted by hydrogen peroxide (H2O2), and it can discriminate Hg2+ through "naked-eye" observation of the color changes from light orange to dark pink. Finally, the practical applications of the probe RH-Hg in the river water further demonstrated that it will be an effective and economical tool for monitoring the distribution of Hg2+ in the environment.
